My gosh, if you put in your job posting that "xyz experience is a plus" but during the interview you tell me you you need an xyz wizard, then whoever started this process just frustrated two people without reason. Yeah I have used that, but not as a big part of my job and certainly not every possible thing that it can do. The company I worked for only used one subset, not the entire suite.
Be clear and honest about what the job actually does every day. Be clear about what you want and need from the perosn you hire. Ambiguity wastes everyone's time.
Oh, and if the hiring manager gives a sigh and an eye roll because you were honest about your skills with said tool, then proceeds to let you know that "this is a senior role and we need someone who needs no training on any facet of this tool", you know you are better off not getting the job.
Hire for fit. Skills can be taught.

An eye roll and sigh from the hiring manager is unprofessional. Red flag to the person being interviewed. If s/he has an issue with the job posting, that is something to be taken up internally, not displayed directly to a candidate.
